Are there significant differences between the rules in the PHB and MM and the stat blocks that were given for modules during the playtest (like Murder in Baldurs Gate)?
For ex. in the Death in Thay module there's a disclaimer:

Depends what you mean by significant tbh.

Looking at the Air Elemental.

A next Air Elemental has AC 13, 45 Hit Points, +5 to hit, 11 Average Damage.
A 5e Air Elemental has AC 15, 90 Hit Points, +8 to hit, 18 Average Damage.

As you can see, the 5e one is quite a bit better. That said, from my play experience Next monsters were pretty much a walk over.

(45 hp vs. 90hp does look significant to me :))

So where are you getting that MM?  Did you convince an all access passholder to give you theirs from Gencon?

The MM won't be out for awhile for most of us.

Best source for 5e monsters for the rest of us is the free download of the DM basic rules here.  

The vast majority of monsters have increased hit dice and points compared to the playtests.

FWIW, another example of how creatures have been beefed up since Dead in Thay.

DiT Basilisk: AC 10, 28 hit points, Bite attack: +4 to hit, 2d6+1 piercing damage.

Basic Rules Updated Basilisk: AC 15, 52 hit points, Bite Attack: +5 to hit, 2d6 +3 piercing damage plus 2d6 poison damage.  (And the save for the gaze attack is DC12 instead of DC 11).

Much more dangerous.

Looks like most of them have been 'scaled up' a few hit dice, so either the playtest creatures could simply be given a lower challenge rating or could be scaled up.
